Methylene Blue Injection USP 1.0% is a sterile injectable solution used for both diagnostic and therapeutic purposes in medical practice. It contains 1% methylene blue, which equals 10 mg of methylene blue per 1 mL of solution.
🔬 Basic Information:Generic Name: Methylene Blue
Strength: 1.0% w/v (10 mg/mL)
Form: Sterile solution for injection
Route of administration: Intravenous (IV)
Pharmacological class: Antidote, Diagnostic dye, Antiseptic (mild), Monoamine oxidase inhibitor (MAOI)
Primary use: Treatment of acquired methemoglobinemia, a condition where hemoglobin is oxidized and cannot carry oxygen efficiently.
Methylene blue acts as a reducing agent, converting methemoglobin back to functional hemoglobin.
Used during surgical procedures as a dye to:
Check tubal patency in gynecology.
Identify fistulas, leaks, or perforations.
Stain tissues for localization.
Used in cyanide poisoning (as part of combination therapy).
Occasionally used in ifosfamide-induced encephalopathy.
In methemoglobinemia, it facilitates the enzymatic reduction of methemoglobin (Fe³⁺) to hemoglobin (Fe²⁺), restoring oxygen-carrying capacity.
As a dye, it stains tissues, fluids, or organs, aiding visual identification during procedures.
Acts as a weak monoamine oxidase inhibitor (MAOI), affecting serotonin levels in rare cases.
Adult dose: 1–2 mg/kg body weight IV over 5 minutes.
A second dose may be administered after 1 hour if needed.
Maximum total dose: usually ≤7 mg/kg.
Must be diluted and administered slowly to avoid side effects like chest pain, hypotension, or confusion.
🧬 Other Notes:Stains tissues, mucosa, and clothing—handle with care.
Urine, stool, and even skin may turn bluish-green temporarily after administration.
Sometimes used off-label for vasoplegic shock or neuroprotective research.
